PCR fluorescence filter is a key component applied in biomedical and life science instruments, mainly used to separate and select the characteristic spectral bands of excitation light and emitted fluorescence of substances in biomedical fluorescence detection and analysis systems. Its working principle is based on the characteristic of fluorescent substances emitting longer wavelength fluorescence under specific wavelength light excitation. By selectively transmitting or reflecting specific wavelength light, it can effectively separate excitation light and emission light, thereby achieving accurate detection of fluorescence signals.
Three sets of core filters elements for PCR optical path
The entire fluorescence PCR optical unit consists of three parts: an excitation bandpass filter, a 45 ° dichroic spectrometer, and an emission bandpass filter. It is responsible for isolating excitation light, purifying fluorescence signals, and eliminating cross color interference.
1. Excitation Filters (Ex Filter, light source side)
Type: Narrowband bandpass BP filter, matched with LED/laser excitation light source, only passing through dye excitation band, blocking light source stray light and ambient background light
Core indicators:
1. Peak transmittance T>85%, FWHM bandwidth 15-30nm (narrow bandwidth reduces channel crosstalk)
2. Cut off depth OD4~OD6 (blocking stray light in the full wavelength range of 200-900nm)
3. Substrate: Optical float glass/quartz, hard dielectric film, temperature resistance -40~120 ℃, suitable for PCR thermal cycling environment
2. Dichroic Mirrors (45 ° spectral core)
Function: Reflective excitation light illuminates the sample downwards and transmits the fluorescence signal upwards to the detector, which is the key to multi-channel PCR beam splitting
Band rule: Reflection band=excitation wavelength ± 20nm (R>95%); Transmittance band=long wavelength band of fluorescence emission (T>95%)
The standard model is named DMxxx, where xxx is the boundary wavelength:
DM505: Reflection 450-490nm, Transmission 515-700nm (FAM channel specific)
DM550: Reflection 450-538nm, Transmission 560-700nm (HEX/Cy3 channel)
DM600: Reflection 450-585nm, Transmission 605-750nm (ROX/Cy5 channel)
3. Emission Filters (Em Filter, detector side)
Type: Narrowband bandpass BP, only releasing target fluorescence, completely blocking reflected strong excitation light (the most critical anti cross color element)
Indicator: OD6 deep blocking excitation band, fluorescence transmission>80%, FWHM 10–25nm， The narrower the multi-channel, the higher the discrimination
The fluorescence PCR filter manufactured by Borisun Optics based on plasma reactive magnetron sputtering (PARMS) process has the characteristics of high transmittance, deep cutoff, and high cutoff steepness. Through in-depth research on the luminescence characteristics of commonly used fluorescent dyes such as Atto 425, FAM, SYBR, VIC, HEX, JOE, TET, TAMRA, Cy3, ROX, Texas Red, CY5, CY5.5, the filter spectrum design was optimized to reduce the cross-talk (cross color) of multi-channel fluorescence detection to an extremely low level, which is widely applicable to real-time fluorescence quantitative PCR (RT qPCR) and digital PCR (dPCR).
