Fluorescence Microscope Filters
A set of falling light fluorescence microscope filter set consists of excitation filter EX, dichroic mirror Dicroic, and emission filter EM, all of which are indispensable:
1. Excitation (EX) of the filter
Position: Between the light source and the dichroic mirror
Function: Filter narrow bands of light sources, only release light that can excite fluorescent dyes, and block other miscellaneous light
Type: Almost all bandpass BP, labeling format: center wavelength/half width, such as 480/30 (465~495nm transparent)
Key indicators: Peak transmittance>85%, cut-off OD ≥ 4 (attenuation of stray light by more than 10000 times)
2. Dichroic dichroic mirror
Installation angle: Fixed 45 ° optical path
Function: Short wave reflection, long wave transmission; Reflect the excitation light towards the sample, and the sample emits longer wavelength fluorescence that directly penetrates the lens
Requirements: steep spectral edges, mirror surface flatness (to avoid image distortion), low wavefront distortion
3. Emission filter (EM/Carrier)
Position: From the two-way mirror to the camera/eyepiece at the last turn
Function: Thoroughly intercept residual excitation light, sample scattered light, and spontaneous fluorescence, and only release target fluorescence signals
Type: Bandpass BP (single dye precision imaging)/Longpass LP (wideband observation)
Definition of Filter Spectral Types
1. BP bandpass (most commonly used): Only through a certain interval, EX and EM mainstream selection
2. LP Long Pass: Transmitting all light with a wavelength greater than the critical wavelength, multi-color imaging, and easy use of fluorescent mirrors
3. SP short circuit: Passing through<critical wavelength, fluorescent devices are rarely used alone

Key technical parameters
1. Transmittance T: The peak transmittance of the working band is ≥ 80%, and the higher the transmittance, the stronger the fluorescence brightness
2. Cut off depth OD
Ordinary laboratory: OD4 (10 ⁻⁴ transparent)
High sensitivity/weak fluorescence, multi-color co dyeing: OD6 (10 ⁻⁶ transparent, no background noise)
3. Steep spectral edge: The narrower the transition interval, the better to reduce dye cross color
